Play in Drive train

Hello,
I am waiting for membership to be approved.
I have an L200 Diamond 2007 auto 108k on the clock.
I have been listening to a quite heaving clunk/knock after I pull up and take the foot off of the brake. This noise was not audible in 4wd. I had the car looked at and the mechanic discovered that the noise appears to be coming from the transfer box. When the truck was on the ramp and the rear wheels turn and are then reversed a clunk can be heard in the transfer box. I have never had a chain driven box before and wondered if this could be a result of a loose chain? Also I not sure why the noise is not there in 4wd.
All help would be appreciated as I have just set myself up in business and desperately need the truck for work and cannot really afford a transfer case replacement.

drop the fluid in the 2 boxes if it has not been done and sieve it see what u find
